Avidin is a biotin-binding tetrameric protein (KD ≈ 1 × 10⁻¹⁴) derived from egg white. It blocks free biotin and biotinylated proteins from interacting with Strep-Tactin® and Strep-Tactin®XT, without affecting Twin-Strep-tag® or Strep-tag®II binding. Because biotin in cell lysates or culture media can reduce resin capacity, avidin should be added to mask biotin before purifying tagged proteins by Strep-Tactin® affinity chromatography.
